Abstract
The aim of this project was to investigate wind tolerant plus trees of Calophyllum inophyllum and then to collect their seeds in Taiwan and Okinawa, Japan. These germinated seeds will be cultivated and seedlings will be planted in coastal area for growth test. Total of 37 plus trees have been investigated in Taiwan, including Hengchung 21 plus trees, Hualien 4 plus trees and Taitung 12 plus trees. Each plus tree with about 100 seeds had been collected, and total 1,243 seedlings have been obtained. Total of 19 plus trees had been investigated in Okinawa, and 177 seedlings were obtained. In 2015, Taiwan seedlings of 32 plus trees and Japan 19 plus trees are also have been planted at Hengchun and Illan sited. In this study, the genetic variation and population structure of 66 individuals sampled from 6 populations of Hernandia nymphiifolia in Taiwan, Green Island and Lanyu were studied using ISSR markers. POPGENE analysis revealed that total gene diversity (H) was 0.1309 and genetic differentiation index (Gst) was 0.5007, while gene flow index (Nm) was 0.2493, which indicated that the genetic differentiation was high among populations. Based on the above finding, both In-situ and Ex-situ conservation are necessary in preserving this species.
