This research project is proposed to develop metabolic profiling approach to the analysis of metabolites based on a simplistic preparation scheme and analysis by electronical ionization-gas chromatography/mass spectrometry. In addition, we will develop the vapor phase extraction technique and use the leaf and callus of Amentotaxus formosana to illustrate metabolic changes in sugars, amino acids and other organic acids. A. formosana are indigenous trees only in the southern parts of Taiwan. A. formosana were propagated using cuttings taken from stump sprouts. The rooting percentage of the cuttings was evaluated after about 4 months. The rooting percentage of some clones was only 24.6% while of others even 86.5%. These trees had already been listed as one of conservative tree species and protected by the government due to their scarce population. Very little effort has been directed toward the chemical composition of A. formosana callus culture. The objectives of this study were to understand chemical component and content of callus induction of A. formosana. Callus were initiated from the leaf excised from A. formosana cuttings. The chemical component of callus were assessed by pyrolysis gas chromatography mass spectrometry(Py-GC-MS).
